Research background: CJC-1295 (No DAC), also known as Modified GRF (1-29), is a synthetic analogue of the first 29 amino acids of endogenous growth hormone-releasing hormone (GHRH), modified for enhanced resistance to enzymatic degradation. Unlike the DAC-conjugated form, it does not bind covalently to serum albumin and is cleared relatively quickly, giving it a pharmacokinetic profile closer to native, pulsatile GHRH signalling. Research has examined its action as a selective agonist of the GHRH receptor on anterior pituitary somatotroph cells, activating the Gs-adenylate cyclase-cAMP-PKA signalling cascade to promote growth hormone gene transcription and pulsatile GH release. It is frequently studied alongside ghrelin receptor agonists such as Ipamorelin to investigate synergistic GH release through complementary receptor pathways.
