Research background: Ipamorelin is a pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) studied as a selective agonist of the growth hormone secretagogue receptor (GHS-R1a) — the same receptor targeted by endogenous ghrelin. Activation of GHS-R1a on pituitary somatotroph cells triggers Gq/11-mediated signalling and intracellular calcium release, promoting GH granule exocytosis. Foundational research identified ipamorelin as notably selective among growth hormone secretagogues: unlike earlier GHRPs such as GHRP-6 and GHRP-2, it did not produce meaningful elevations in ACTH, cortisol, prolactin, FSH, LH, or TSH, even at doses substantially above its GH-releasing ED50. This selectivity profile is why it is frequently studied alongside GHRH analogues such as CJC-1295 to investigate complementary receptor pathways in GH release.
